cpnA cells have unusually large contractile vacuoles when placed in water. (A) Wild-type (WT) and cpnA (cpnA-) cells in HL-5 were placed in glass-bottom dishes and allowed to settle. The HL-5 was replaced with water, and after 2 h, the cells were imaged using DIC microscopy. (B) Wild-type and cpnA clones in HL-5 were placed in glass-bottom dishes and allowed to settle. The HL-5 was replaced with water and the numbers of cells with vacuoles larger than one-fourth of the area of the cell were counted at various time points using DIC microscopy. The percentages of cells containing large vacuoles from three independent experiments were calculated and then averaged. The error bars represent standard error.
