Transcriptional activation is restored by increasing amounts of Smad2(R133C) but not Smad4(R100T). HepG2 cells were transfected with the gsc-Lux reporter alone or together with increasing amounts of wild-type (wt) or mutant versions of Smad2 or Smad4 with (+) or without (−) FAST2, as in Fig. 1.
