Cys246Ala mutation in the C1 domain of β2-chimaerin abolishes [3H]PDBu binding. COS-1 cells were transfected with pEGFP, pEGFP-β2-chimaerin, or pEGFP-C246A-β2-chimaerin. After 48 h binding of [3H]PDBu was measured in total cellular lysates by using 15 nM radioligand, as described in Materials and Methods. Two additional experiments gave similar results.
