In the absence of Oye2p, actin oxidation activates the oxidative stress response. The oxidative stress responsive transcription factor Yap1p was expressed as a GFP fusion in various mutants and the percentages of cells with nuclear GFP-Yap1p was quantified by fluorescence microscopy.
