Cardiomyocytes derived from MyHC-ATF3 transgenic ventricles have decreased contractile function. Ventricular cardiomyocytes were isolated from NTG, TG1, or TG2 mice at 15 to 20 weeks of age. Their percentage of cell shortening, lusitropy, and inotropy were measured after stimulation in the electrical field in the absence or presence of isoproterenol. Percent of cell shortening = (L0 − Li)/L0, where L0 is the length of resting cells and Li is the length of the electrically stimulated cells. There was no statistically significant difference in the resting cell length between TG and NTG cells: NTG 104 ± 3 μm (n = 26), TG1 101 ± 5 μm (n = 24), TG2 109 ± 4 μm (n = 28).
