The HRA1 gene in S. cerevisiae Chromosome 1. (A) A cloned HRA1 gene is located in yeast chromosome I from 99,306 to 99,869 in the antisense strand of a known coding region for DRS2 (YAL026C). The upstream sense strand MAK16 (YAL025C) runs from 101,146 to 100,226 nt. (B) A yeast HRA1 gene was amplified directly from intact yeast cells by using primer oligonucleotides HRA1F3 (5′-TAATACGACTCACTATAGGCCCTTTCTTCCGTTTGAA-3′) with T7 promoter sequence (in italics) and HRA1R3 (5′-CATATGTTTGCCAGAAAACCCAAATC-3′) with the NdeI site (underlined). The overlapped sequence with the DRS2 gene is shown in bold letters. The sequence of the cloned HRA1 gene was used for WU-BLAST2 analysis with S. cerevisiae genomic sequence (http://seq.yeastgenome.org/cgi-bin/nph-blast2sgd) and shows 99% homology with the yeast genomic sequence.
