Atp9 assembles into an F₁-containing subcomplex in the absence of Oxa1. Mitochondria from wild-type (WT; A), Δoxa1 (B), Δatp10 (C), and rho⁰ (D) strains were solubilized with 1% Triton X-100 containing buffer and subjected to size exclusion chromatography on Superose 12 column. Fractions (11–27) eluted from the column were TCA precipitated and subjected to SDS-PAGE followed by Western blotting with antisera against F₁ subunit Atpβ and F_o subunits Atp6 and Atp9. The elution profile of the molecular mass standards 443, 200, 150, 66, and 29 kDa are indicated in the wild-type panel. Note the exposure time for Atp6 in the Δoxa1 and Δatp10 mitochondria was significantly longer than that for the wild-type control, because of the strongly decreased levels of Atp6 in these mutant mitochondria.
