Stimulation of 125I-α2-macroglobulin uptake and degradation in granulosa cells by Bt2cAMP plus hLDL. Bt2cAMP-sensitized granulosa cells were treated with Bt2cAMP (2.5 mM) plus hLDL (100 μg/ml; open triangles) or maintained in medium containing Bt2cAMP (2.5 mM; open circles) for 24 h as described in Experimental Procedures. They were then incubated with 125I-α2-macroglobulin (1 μg/ml) for 60–240 min at 37°C. A: After washing as described, the amount of 125I-α2-macroglobulin associated with the cells (total uptake) was determined by dissolving the cells with 0.1 N NaOH. B: Degradation was measured as the radioactivity in the incubation medium that was soluble in 15% trichloroacetic acid. Results are of duplicate determinations. The extent of variation among the data points was between 7.8% and 14.0%.
