Only certain laboratory strains of HSV trigger activation through TLR2. (A) HEK293T cells expressing TLR2, CD14, and NFκB-driven firefly luciferase reporter gene (293T/luc/TLR2/CD14 cells) were cultured with the indicated MOI equivalents (MOIe) of UV-inactivated HSV-1 KOS strains from three different sources (A, ATCC; K, Knipe; CE, Cohen and Eisenberg) or 10 μg/ml zymosan for 7 h, and luciferase activities were measured. (B) 293T/luc/TLR2/CD14 cells were cultured with the indicated MOIe of UV-inactivated HSV-1 or HSV-2 strain, zymosan (10 μg/ml), or 0.02% SAC for 7 h, and luciferase activities were measured.
