Reverse transcription initiation efficiency of the wt and J1–J10 transcripts. (A) DNA or tRNAlys3 primers were annealed to wt and mutant 1-368 transcripts and extended with 32P-CTP by the HIV-1 RT enzyme. Extended primers were separated on a denaturing acrylamide gel. The 1/2 tRNA signal represents a half tRNA species that is discussed in detail in the context of Figure 8. (B) Relative initiation of reverse transcription efficiency of the wt and mutant transcripts, the wt activity was arbitrarily set at 1. The average and standard deviations of three independent experiments were quantified.
