Groups of mice (n = 5) were inoculated by gavage with 2 × 107 CFU of type A F. tularensis strain FSC033 at day 0, and the MLN, liver, and spleen were collected from each animal at 0, 1, 2, and 3 dpi. The relative levels of cytokine and chemokine mRNA expression were determined by real-time PCR analysis as described in Materials and Methods. Mouse β2-microglobulin RNA was measured and used to calculate relative expression using the formula Rel Exp = 2ΔΔCt. The results shown are the average of relative expression values determined using cDNA from three mice, with ranges indicated in parentheses.
