Southern blots of transient DNA replication supported by UL42 mutant. The transient DNA replication of the oriS-containing plasmid (pHOS9.2) supported by the UL42 mutants was performed as described in Materials and Methods. Aliquots of purified DNA samples were digested with either EcoRII alone or with EcoRII plus DpnII, fractionated on a 0.8% agarose gel, and hybridized to a probe corresponding to the ColE1 sequence. (A) Results shown from the wild-type UL42 control (lanes 1 and 2) and the mutation R279A (lanes 3 and 4).
