Lytic DNA replication is SM dependent. SM-KO EBV-infected 293 cells were transfected with either vector plasmid (C), Z, or Z plus SM (Z/SM) and analyzed by Gardella gel electrophoresis 48 h posttransfection. EBV DNA was detected by Southern blotting with radiolabeled BamHI W fragment of EBV DNA. Untreated B95-8 cells (−TPA) and B95-8 cells induced to replicate lytically by treatment with TPA (+TPA) were used as controls. L, linear genomes.
