Single-copy plasmids expressing Mks1p from its native promoter were used to obtain a modest increase in expression of this protein. Transformants of YHE869 (MATα ura2 leu2Δ∷hisG trp1Δ∷hisG [ure-o]) with the indicated plasmids were grown without tryptophan or leucine and suspended in water, and dilutions were plated on SD + USA. Number of colonies formed after 5 days is shown. Plasmids: pH7 = 2 μ LEU2 PADH1, pH14 = 2 μ LEU2 PADH1URE2, pH438 = 2 μ LEU2 PADH1URE2ΔApaI, pH417 = CEN TRP1 PMKS1, pH431 = CEN TRP1 PMKS1MKS1. The experiment was carried out four times with similar results; typical data are shown.
