Combined immunoprecipitation and immunoblotting analysis of KS-bearing and KS-free glycoforms of thyroglobulin and transferrin produced by PTC. Material extracted from PTC lesions as described for Figure 2 was immunoprecipitated by the first listed antibody, resolved by SDS-PAGE under nonreducing conditions in gradient gels, as indicated on the top (ie, 3 to 15%, 3 to 12%, and 3 to 8% gradients), electrotransferred onto nitrocellulose or PDVF membranes, and immunoblotted with the second antibody (this procedure referred to with a +). Alternatively, the material was immunoprecipitated with as above and the supernatant collected, concentrated, resolved by SDS-PAGE, and blotted with the heterologous antibody. The 373E1 IgM served as the 900-kd Mr marker. AsTG, anti-thyroglobulin antiserum; AsTF, anti-transferrin antiserum.
