Target nucleotide methylation requires a continuous, Watson–Crick paired guide:target RNA duplex. D target RNAs possessing mismatched nucleotides were incubated with in vitro assembled sR8 box C/D sRNP and 2′-O-methylation of the target nucleotide assessed.
