Raising the level of TFIIIC2 stimulates pol III transcription in
ovarian epithelial cell extracts. (A) In vitro
transcription by using 10 μg of ROSE 199 ovarian cell extract and 250
ng of pVA1 template. Reactions 2, 3, and 4 were supplemented with 2, 4,
and 8 μl of phosphocellulose B, respectively. Reactions 6, 7, and 8
were supplemented with 2, 4, and 8 μl of phosphocellulose C,
respectively. (B) In vitro transcription
assay by using 10 μg of ROSE 199 ovarian cell extract and 250 ng of
pVA1 template. Reactions 2, 3, and 4 were supplemented with 3, 6, and 9
μl, respectively, of TFIIIC that had been immunopurified by using
antiserum 4286 against TFIIIC110. Reactions 5, 6, and 7 were
supplemented with 3, 6, and 9 μl, respectively, of material that had
been mock immunopurified by using preimmune serum.
