TrkB increases the half-life of Kv1.3 channel protein. (A) HEK293 cells were transiently transfected with cDNA encoding Kv1.3 (K) or Kv1.3 plus TrkB (KT). Twenty-eight hours following transfection, cells were treated with two different concentrations of cycloheximide and then lysates were harvested at varying time intervals (in hours) as noted. Lysates were separated by SDS PAGE and nitrocellulose was blotted with anti-Kv1.3 as in Fig. 1. (B) Line graph of the normalized immunodensity values for the Kv1.3 band versus time following 100 μg/ml cycloheximide treatment as in (A).
