K263 is a critical residue for centrosomal localization of NPM/B23. NIH 3T3 cells were transfected with either FLAG-wild-type NPM/B23 (e–h) or FLAG-K263R mutant (i–l). The vector plasmid was transfected as a control (a–d). At 24 h posttransfection, cells were fixed with methanol and coimmunostained with anti-FLAG and anti-γ-tubulin antibodies.
