(A) Effects of voltage on KIFMK block of F1489Q currents. Macroscopic currents were elicited from excised inside-out patches of membrane expressing the F1489Q channels in the presence of 100 μM KIFMK by voltage steps from −100 mV to a range between −30 and 30 mV in increments of 10 mV. Also shown are the superimposed single exponential fits of Eq. 1: y(t) = A * exp(t/τ) + B (see materials and methods) to each decaying current trace. (B) Effects of voltage on the rates of KIFMK block of F1489Q currents. Plot shows the on (•) and off (○) rates of 100 μM KIFMK as a function of voltage. The smooth lines represent a fit of the equation k b = 1,390*exp(V/43 mV) to the blocking rates. The unblocking rates showed no voltage dependence (○).
