Subcellular localization and kinase activity of FRAP proteins tagged
with various translocation signals. (A) Myc-tagged NLS-,
NLS′-, NES-, and NES′-FRAP proteins were transiently expressed in CV-1
cells, followed by immunostaining and confocal microscopic analysis.
(B) The same recombinant FRAP proteins were transiently
expressed in HEK293 cells and immunoprecipitated by an anti-Myc
antibody, and in vitro autokinase assays were performed
in the immune complexes. All FRAP constructs, including wild-type,
contained the S2035T mutation.
