Cav-3 expression does not affect the detergent solubility of two other PFK isoforms, namely PFK-B and PFK-P. Cos-7 cells were transiently transfected either with the cDNA encoding V5-tagged PFK-B, alone or in combination with Cav-3 (A) or with the cDNA encoding V5-tagged PFK-P, alone or in combination with Cav-3 (B). Thirty-six hours after transfection, the soluble fraction was extracted by incubating cells with a buffer containing 1% Triton X-100. The insoluble fraction was collected using 1% SDS. Equal volumes of the soluble (S) and insoluble (I) fractions were resolved by SDS-PAGE and analyzed by immunoblotting with V5 or Cav-3 mAb probes. Note that Cav-3 expression does not affect the detergent solubility of the other two PFK isoforms, ie, PFK-B and PFK-P. PFK-B partitions both in the soluble and in the insoluble fractions, either when expressed alone or in combination with Cav-3 (A). B shows that PFK-P is mainly found in the insoluble fraction; the presence of Cav-3 does not affect this behavior.
