h-Int6 and Moe1 interaction is conserved during evolution. (A) yin6Δ cells (YIN6K), transformed with either a vector control (pSLF173) or various plasmids (pHAHINT6, pHAT1139, pHAYIN6, and pHAT22) carrying the indicated genes, were spread on MM medium containing 0, 0.2, 2, or 20 μM thiamin at 23°C. We obtained the same results regardless of the thiamin concentration tested, and show herein only those with 20 μM. On the right is a Western blot showing the expression of full-length and truncated h-Int6 in wild-type cells. (B) Protein–protein interactions as tested by the yeast two-hybrid system. The β-galactosidase activity (Miller units) was measured on three independent colonies per transformation. The plasmids tested were pGADGH, pGADHINT6, pGADT1139, pGADT22, pGADYIN6, pVJL11, pLBDMOE1, and pLBDHMOE1.
