Ama1p associates with the APC/C in vivo. (A) Extracts were prepared from a meiotic timecourse of a wild-type diploid harboring chromosomally tagged AMA1-HA and CDC16-myc. Ama1p-HA (Upper) and Cdc16-myc (Lower) accumulation in these extracts was determined by immunoprecipitation and Western blot analysis. Meiosis I and II were completed by 9 and 12 h, respectively. (B) Ama1p associates with Cdc16p in vivo. The Cdc16p-myc blot shown in A was stripped and probed for Ama1p-HA. Ama1p-specific bands are indicated by the arrows (Upper).
