Hoxa10 expression in relation to osteogenic differentiation. (A) Premyogenic C2C12 cells were treated with 100 ng/ml BMP2 for 24 h. Total RNA was isolated at different time points (0, 1, 2, 4, 6, 8, 12, 16, 20, and 24 h). Five micrograms of total RNA was reverse transcribed with oligo(dT) primer and amplified (RT-QPCR) by gene-specific primers. (Top) Hoxa10 expression (+BMP, −BMP) was normalized to Gapdh expression (+BMP, −BMP), and relative transcript levels were plotted; (bottom) the induction of Runx2 expression with BMP2 treatment is shown for the same time course. Independent experiments exhibited similar temporal expressions (data not shown).
