Complement is activated during RRV infection. Twenty-four-day-old C57BL/6J wild-type, C3−/−, or RAG-1−/− mice were infected with 103 PFU of RRV by injection in the left rear footpad. At 7 dpi, mice were euthanized, serum was collected, mice were perfused with 1× PBS, and the left ankle or quadriceps muscle was homogenized in radioimmunoprecipitation assay lysis buffer. Immunoblot analyses were performed with polyclonal anti-murine C3 antibody (Cappel) or an anti-α-tubulin antibody as described in Materials and Methods.
