Effects of TXA2 receptor agonist or antagonists on mRNA expression and the production of MCP-1 in human umbilical vein endothelial cells (HUVEC). (a) HUVEC were not stimulated (lane 1), or stimulated with U46619 (10−7 m or 10−6 m) (lanes 2 and 3) and incubated for 6 h. SQ29 548 (10−5 m) or BAYu3405 (10−5 m) were pretreated before the addition of 10−6 m U46619 (lanes 4 and 5). Reverse transcription-polymerase chain reaction (RT-PCR) was done with specific primers for MCP-1 or β-actin on mRNA extracted from 107 cells as described in Materials and Methods. Shown is a gel photograph representative of three similar experiments. (b) The PCR products were quantified by quantitative image analysis as the ratio in relation to β-actin mRNA (1·0). (c) The production of MCP-1 was determined as the concentrations of MCP-1 in the culture supernatants after 24 h stimulation using ELISA.
