Effects of costimulation on Th2 cytokine production by human T cells. One million T cells were cultured with 106 mitomycin C-treated wild-type P815 cells or P815 cells transfected with CD80 or CD86 in a final volume of 1 ml of complete medium in flat-bottomed 24-well culture plates. Anti-CD3 MoAb UCHT1 (2 μ g/ml) was used as the primary stimulus. After 1 day (IL-4) or 4 days (IL-5, IL-13), supernatants were collected and cytokines were measured by ELISA. The results are from three independent experiments.
