Th2 cytokine production by CD4+ and CD8+ T cells. One million CD4+ or CD8+ T cells isolated from blood of five (a) or 10 (b,c) different donors were cultured with 106 mitomycin C-treated P815/CD80 cells in a final volume of 1 ml complete medium in flat-bottomed 24-well plates. Anti-CD3 MoAb UCHT1 (2 μ g/ml) was used as the primary stimulus. For the cultures of (a), anti-IL-4R MoAb (2·5 μ g/ml) was added to block IL-4 consumption. Supernatants were collected after 4 days of culture. The concentrations of IL-4, IL-5 and IL-13 were determined by ELISA.
