Effects of calcium rise on Th2 cytokine production. One million T lymphocytes were incubated with 10⁶ P815 or P815/CD80 cells in a final volume of 1 ml complete medium in flat-bottomed 24-well plates. Phorbol myristate acetate (PMA; 1 ng/ml) and/or ionomycin (0·5 μ g/ml) were added as primary signals. Supernatants were taken at day 1 (IL-2, IL-4) or at day 4 (IL-5, IL-13). Cytokine levels were determined by ELISA. The figure shows the results of independent experiments on T cells from eight to 19 donors.
