Major basic protein (MBP) induces IL-8 mRNA levels in human intestinal myofibroblasts. 18-Co human intestinal fibroblasts were incubated for 3 h with MBP (10⁻⁶ m) or medium alone. Total RNA was isolated and reverse transcription-polymerase chain reaction performed using primers specific for human IL-8. Southern blot analysis was performed with a ³²P-labelled IL-8 cDNA probe. Blots were stripped and re-probed with a chicken glyceraldehyde 3-phosphate dehydrogenase (GAPDH) cDNA probe to assess RNA loading.
