Competition of low molecular weight molecules with GTP-TR for formation of complexes with α₂M*. A 50-fold molar excess (1mM) of competing molecule was co-incubated with GTP-TR (20μM) and non-proteolytically activated α₂M* for 5h at 50°C in 25mM HEPES pH 8.0, 200mM NaCl following which unbound ligand was separated and the fluorescence of the α₂M* complex was quantified. A decrease in fluorescence indicates competition of the low molecular weight molecule with GTP-TR for formation of a complex with α₂M*. All data points are the mean ± 1 SEM, with n=6.
