Endosomal maturation/acidification is required for CpG ODN 1826 to activate NO and iNOS production. RAW 264·7 macrophages (1 × 106 cells/well) were preincubated with or without chloroquine (2·5 μg/ml) for 2 h before activated with CpG ODN 1826 (1 μg/ml) or E. coli LPS (10 ng/ml). After 24 h, the supernatant was analysed for nitrite (a) and the cells were analysed for iNOS (b). Control represents the unstimulated cells.
