Protein levels reflect the Pip2p-independent role of Oaf1p as a negative regulator. Wild-type and deletion strains each with a GFP-tagged version of an AOY cluster gene (DIP5 or ATO3) were analyzed by FACS in the presence of glycerol and 20 h (Dip5-GFP strains) or 5 h (Ato3-GFP strains) after the addition of oleate to the medium. Deletions of OAF1 but not PIP2 resulted in lower levels of fluorescence relative to wild-type levels. Deletion of ADR1 or OAF3 resulted in decreased or increased Dip5-GFP levels, respectively.
