(A) Identification of the transcription start site. The RNA transcribed from the genes containing the upstream sequences of TTHB186 (TTHB186_up), TTHB178 (TTHB178_up), TTHB147 (TTHB147_up), TTHA0771 (TTHA0771_up), TTHA0176 (TTHA0176_up), and TTHB159 (TTHB159_up), respectively, in the presence of both T. thermophilus CRP and cAMP, was reverse transcribed with avian myeloblastosis virus reverse transcriptase (lane 5), as described in Materials and Methods. The nucleotide sequence of the template DNA was determined by the dideoxy-mediated chain termination method (lanes 1 to 4). After the reaction, samples were analyzed by PAGE, followed by autoradiography. The 3′ terminus of the cDNA is indicated by an asterisk.
