Step-by-step efficiency of cloning, small-scale protein expression and purification of E. coliLUCA proteins. 177 purified proteins with high and moderate yields were subjected to large-scale expression. 142 proteins could be purified with a minimum of 500 μg. 32 target genes were subjected to Gateway recombination cloning in order to express fusion proteins with MBP-His7, GST-His7, and NusA-His6, respectively. Large-scale expression revealed 19 purified proteins fused to MBP-His7 with a minimum of 500 μg.
