Transgene expression from liver cells transduced with AAV2 vectors in the presence of AAV2 capsid-specific CTLs. Spleen cells from immunized mice were further stimulated in vitro with DCs for 5 days (Materials and Methods). A total of 2 × 1011 particles of rAAV2 were injected into the liver via the portal vein 30 min prior to tail vein infusion of activated spleen cells. The Control group represents the group without the infusion of AAV2 capsid-specific CTLs, the CTL-AAV2 group represents the group in which the CTLs were induced by DCs pulsed with AAV2 vectors, and the CTL/Adcap2 group represents the group in which CTLs were induced by DCs transduced by Ad/cap2 vectors. Transgene expression was measured at the indicated time points. The data represent the average ± standard deviation from five mice. (A) Liver transduced with AAV2/cap2-gluc.
