Synthesis of SIVcpzPts proviruses from fecal RNA consensus sequences. The complete TAN2 and TAN3 genomes were amplified as partially overlapping RT-PCR fragments (open boxes) from fecal virion RNA and sequenced without interim cloning (fragments are drawn to scale). The resulting population sequences were used to synthesize the TAN2 and TAN3 genomes as proviral halves (black lines). Restriction enzymes introduced into the sequence allowed their directional cloning into an appropriate plasmid vector (pCR-XL TOPO).
