Glideosome solubility in Triton-X100 is temperature and buffer dependent. Parasites were harvested and subjected to detergent extraction in either 1%TX100 in PBS or 1%TX100 in TBS. Extractions were performed for 10 min at 37°C or on ice. Extracts were subjected to centrifugation for 10 min at 14,000 × g and 4°C. Proteins in the total lysate (TOT), supernatant (SN), and pellet (P) fractions were resolved by SDS-PAGE, transferred to nitrocellulose, and incubated with antisera to glideosome proteins and the membrane skeleton protein IMC1.
