Figure 4. — Heterologous DLC-ASs are able to confer interaction with DLC to fusion proteins. Cells were transfected and analyzed by CLSM as described in the legend to Figure 2. (A) GFP-p53BP1_1117-1177 and GFP-PTHR_478-511 show subcellular distribution that is not different from GFP alone (single transfection, compare with Figure 2A). In cotransfected cells, GFP-p53BP1_1117-1177, but not GFP-PTHR_478-511, colocalizes with dsRed-LC8 in the cytoplasm (dual transfection). (B) GFP-DLC-AS fusion proteins containing the T-agNLS are localized to the nucleus of transfected cells. dsRed-LC8 protein colocalizes in the nucleus with GFP-RPP_139-174-T-agNLS and GFP-p53BP1_1117-1177-T-agNLS (middle panel), but not GFP-T-agNLS, GFP-PTHR_478-511-T-agNLS, or GFP-RPP_139-174(D₁₄₃/Q₁₄₇-A)-T-agNLS (top and bottom panels).
