Depletion of Utp10 and Utp20. (A) Growth rate of wild-type, GAL∷HA-UTP10, and GAL∷ProtA-UTP20 strains following a transfer from permissive galactose medium to nonpermissive glucose medium for the times indicated. Cells were maintained in exponential growth throughout the time course by addition of pre-warmed medium. (B) Western analysis of 3HA-Utp10 and ProtA-Utp20 depletion. Total protein was extracted at the times indicated and analyzed by Western blotting with antibodies that decorate the tags present on Utp10 and Utp20, or with anti-Nop1 as loading control. 3HA-Utp10 was decorated with rabbit anti-HA primary antibody, which was subsequently detected using anti-rabbit IgG linked to horseradish peroxidase. ProtA-Utp20 was detected with a peroxidase-antiperoxidase (PAP) complex.
