Vesicle budding reactions were performed at 30°C with microsomes prepared from strains bearing sec16 alleles. Release of the five assayed molecules was normalized to the wild-type values obtained under the same conditions. Error bars are indicated, based on two repetitions of the reactions. In these experiments, the wild-type values were as follows: 29% gpαF, 23.3% Sec22p, 8.5% Bos1p, 14.8% Bet1p, and 7.5% Emp24p. There was no detectable release of Emp24p from the sec16-2 microsomes.
