Cell extracts were applied to SDS-PAGE for Western blotting with anti-IκBα and anti-β-actin antibodies. (B–D). Adr induces the formation of IKK and RIP complex. Cell extracts were prepared from mouse wild-type (B), TRAF2−/− (C), and ATM−/− cells (D) after being treated with Adr (10 μg/mL) for various times as indicated. Cell extract from each sample was immune-precipitated with an anti-IKK-γ antibody overnight. Immune-precipitants were analyzed by Western blotting with anti-RIP, anti-TRADD, and anti-IKK-β antibodies. One percent of cell extract from each sample was used as a control of protein input.
