The three major states of pHLIP at concentration of < 30 μg/mL are illustrated: soluble in water at pH > 7 (State I), bound to the surface of a lipid bilayer at the same pH and at a lipid/peptide molar ratio > 100 (State II), and inserted across the bilayer as an α-helix at low pH (State III).
