Nuclear extracts prepared on the days indicated were subjected to Western blot analysis or EMSA using the CUP-1 binding site oligonucleotide as labeled probe. D-4 refers to proliferating 3T3-L1 preadipocytes; D-2 and D0 refer to confluent and 2-day postconfluent preadipocytes; and D1, D2, D3, D4, D5, D6, and D7 refer to cells 1, 2, 3, 4, 5, 6, and 7 days, respectively, after induction of differentiation. (B) AP-2α protein was detected by Western blot analysis of nuclear extracts during differentiation of 3T3-L1 preadipocytes.
