G4 stabilization by RHPS4 can induce short-term telomere damage and release of POT1. Overexpression of either TRF2 or POT1 can prevent G4 formation, for example, by favoring t-loop formation or G4 unwinding, and render cells resistant to RHPS4-induced damage. RHPS4 can also induce long-term effects through telomerase inhibition and consequent telomere shortening.
