Effect of FTY720 on IL-1β-induced protein secretion and activity of sPLA₂ in mesangial cells. Quiescent mesangial cells were stimulated for 24 h with either vehicle (−) or interleukin-1β (IL-1β, 1 nM; +) in the absence or presence of the indicated concentrations of FTY720 or FTY-phosphate (pFTY; both in μM). Thereafter, supernatants were taken for protein precipitation followed by protein separation by SDS-PAGE, transfer to nitrocellulose and Western blot analysis using a specific monoclonal antibody against rat IIA-sPLA₂ antibody at a dilution of 1 : 60 (a), or sPLA₂ activity was measured in the supernatants as described in the Methods section (b). Data in (a) show one representative blot out of at least three independent experiments. Data in (b) are expressed as % of maximal IL-1β-induced activity and are means±s.d. (n=3), ^**P<0.01, ^***P<0.001 considered statistically significant when compared to the IL-1β-stimulated values.
