Saturation assay of [³H]5HT binding to 5HT2c receptors expressed in HeLa cells in the absence (○) or presence (•) of EEDQ. At room temperature (≈22°C), EEDQ pretreatment caused a 40% decrease in the binding B_max, without significantly altering the K_d: without EEDQ, the K_d and B_max were 10.1 nM and 4.2 pmol/mg protein; with EEDQ, 14.7 nM and 2.6 pmol/mg of protein, respectively. Similar results were obtained in two other experiments. In this and the following figures, unless otherwise stated, EEDQ treatment refers to incubating the membranes with 2 × 10⁻⁴ M EEDQ for 1 h.
