TcRBP19 is located in the T. cruzi amastigote cytoplasm. Localization was performed as described previously (Dallagiovanna et al., 2005). Parasites were harvested from culture media, washed four times with PBS and resuspended (5 × 106 cells/mL). Fixed cells (4% paraformaldehyde in PBS) were blocked with PBS-Tween, BSA 1% for 30 min., incubated with anti-GST-TcRBP19 for one hour and then with antirabbit Alexa-488 F(ab') fragment of goat antirabbit IgG (H+L) (Molecular probes). Counterstaining was done using propidium iodide. Images were obtained using a FV300 Olympus Confocal microscope using a 100x lens (oil, N.A.1.35) with Argon (488nm) and HeNe (546nm) laser excitation.
