Wild-type cells (WT, RH448) and lcb1-100 (lcb1, RH3802) cells carrying either a YCK2 or a YCK1 high copy number plasmid (lcb1+YCK2 or lcb1+YCK1) were assayed for LY accumulation in the vacuole at 37°C. The same field of cells viewed by fluorescence (left panels) and by Nomarski optics (right panels) is shown. Note that lcb1-100 cells have fragmented vacuoles when compared with wild-type cells (right panels).
